TheGENOMESUNCOUPLED1(GUN1)genehasbeenreportedtoencodeachloroplast-localizedpentatricopeptide-repeatprotein,whichactstointegratemultipleindicatorsofplastiddevelopmentalstageandalteredplastidfunction,aspartofchloroplast-to-nucleusretrogradecommunication.However,themolecularmechanismsunderlyingsignalintegrationbyGUN1haveremainedelusive,upuntiltherecentidentificationofasetofGUN1-interactingproteins,byco-immunoprecipitationandmass-spectrometricanalyses,aswellasprotein–proteininteractionassays.Here,wereviewthemolecularfunctionsofthedifferentGUN1partnersandproposeamajorroleforGUN1ascoordinatorofchloroplasttranslation,proteinimport,andproteindegradation.Thisregulatoryroleisimplementedthroughproteinsthat,inmostcases,arepartofmultimericproteincomplexesandwhoseprecisefunctionsvarydependingontheirassociationstates.Withinthisframework,GUN1mayactasaplatformtopromotespecificfunctionsbybringingtheinteractingenzymesintocloseproximitywiththeirsubstrates,ormayinhibitprocessesbysequesteringparticularpoolsofspecificinteractors.Furthermore,theinteractionsofGUN1withenzymesofthetetrapyrrolebiosynthesis(TPB)pathwaysupporttheinvolvementoftetrapyrrolesassignalingmoleculesinretrogradecommunication.
