Callus cultures of two susceptible Castanea sativa cultivars, ‘Garrone rosso’ and ‘Clone 71’ were grown on culture medium supplemented to 50% with culture filtrates (CFs) from E4 virulent (E4-V) or E13 hypovirulent (E13-H) Cryphonectria parasitica strain, respectively. E13-H CFs caused a reduction in fresh weight and an increase in dry weight on calli of both genotypes. E4-V CFs did not induce any modification of these parameters compared with the control, but fresh weight of ‘Clone 71’ was reduced. Light and electron microscopy observation showed the presence of large osmiophilic aggregates in the vacuoles and, sometimes, the ruptured tonoplast in the E4-V treated calli. Calli grown on El 3-H CFs had a meristematic-like appearance, with small, scarcely vacuolated cells containing altered mitochondria and plasmalemma proliferations.

Abstract

Modifications de la croissance et de Vultrastructure des cals de chdtaignier induites par des filtrats de cultures de souches virulentes ou hypovirulentes de Cryphonectria parasitica Des cals de deux cultivars sensibles de Castanea sativa, 'Garrone rosso' et 'Glone 71', ont ete cultives sur un milieu supplemente a 50% avec des filtrats de culture (EGs) de la souche virulente E4 (E4-V) ou de l'hypovirulente E13 (E13-H) de Cryphonectria parasitica. Ees EGs de E13-H provoquaient une reduction du poids frais et une augmentation du poids sec des cals des deux genotypes. Les FGs de E4- V n'lnduisaient aucune modification de ces parametres par rapport aux temoins, a I'exception du 'Glone71' dont le poids frais etait reduit. Ees observations en microscopie optique et electronique ont permis de detecter chez les cals de E4-V traites aux EGs, de nombreux agregats osmophiles dans les vacuoles et parfois, la rupture du tonoplaste. Ees cals cultives en presence des EGs de E13-H presentaient un aspect semblable a celui d'un meristeme, caracterise par des cellules petites, peu vacuolisees, contenant des mitochondries alterees et des proliferations du plasmalemme.
Wirkung von Kulturfiltraten virulenter und hypovirulenter Stamme von Cryphonectria parasitica auf das Wachstum und die Ultrastruktur von Castanea sativa-Kallus Kallus zweier anfalliger Castanea sativa-Genotypen ('Garrone Rosso' und 'Glone 71') wurde auf Medien kultiviert, die mit 50% Kulturfiltrat von virulenten (E4-V) beziehungsweise hypovirulenten (E13-H) Cryphonectria parasitica-Stammen versetzt waren. E13-H verusachte eine Reduktion des Erischgewichts und eine Zunahme des Trockengewichts beider G. sativa-Genotyyen. E4-V veranderte dagegen nur das Erischgewicht des C. sativa Genotyps 'Glone 71'. Eicht- und elektronenmikroskopische Aufnahmen zeigten, dafi E4-V zu zytopathischen Veranderungen im Kallus von C sativa fiihrte (Auftreten grofier osmiophiler Aggregate in den Vakuolen sowie zum Teil Zerstorung der Tonoplasten). Atif E13-H gewachsene C sativa-Y^sWi wiesen dagegen eine meristematische Grundstruktur auf. Die kleinen, nur wenig vakuolisierten Zellen zeigten eine veranderte Produktion von Mitochondrien und Plasmalemma.
