Functional feed additives with antibacterial and antioxidant properties play a crucial role in countering infections, enhancing animal health, and optimizing host functions. This PhD project aimed to evaluate innovative functional feed additives (algae, tannin extract, leonardite, tributyrin, and probiotics) as alternatives to antibiotics, focusing on their impact on health, performance in weaned piglets through in vitro, in vivo, and ex vivo approaches. In vitro methods characterized additives for antimicrobial and antioxidant effects, while cell cultures predicted their interaction with the intestinal epithelium. In vivo trials assessed additives effect on health and performance in weaned piglets. Results revealed multiple bioactive properties, positive impacts on performance, reduced diarrhoea occurrence, improved health status, and microbiota modulation. Not all additives consistently boosted growth performance in vivo, though they notably reduced gastrointestinal disorders. Lastly, an ex vivo swine intestinal perfusion model was developed to study nutrient bioaccessibility, aligning with the 3R principles. Dietary approaches with functional feed additives significantly contribute to decreasing pathology occurrence, enhancing animal health and performance, thereby reducing antibiotic use. The use of functional feed additives presents a valuable alternative by preventing multifactorial disorders in swine farming, in line with One Health principles.
Gli additivi funzionali agiscono positivamente nei confronti di infezioni, salute e performance degli animali. Questo lavoro di tesi di dottorato ha avuto come obiettivo quello di valutare additivi funzionali innovativi (alghe, estratto di tannini, leonardite, tributirrina e probiotici) come alternative agli antibiotici, valutando il loro impatto sulla salute e crescita dei suinetti svezzati attraverso approcci in vitro, in vivo ed ex vivo. I metodi in vitro hanno permesso di caratterizzare gli additivi per i loro effetti antimicrobici, antiossidanti, e la loro interazione con le cellule intestinali. Gli studi in vivo hanno valutato l’effetto sulla salute, crescita e modulazione del microbiota in suinetti svezzati. I risultati hanno rivelato molteplici proprietà bioattive degli additivi, impatti positivi sulle performance, riduzione dell’incidenza di diarrea e modulazione del microbiota. Non tutti i composti hanno migliorato le performance, sebbene abbiano ridotto i disturbi gastrointestinali. Infine, è stato sviluppato un modello di intestinale di suino riperfuso per studiare la bioaccessibilità dei nutrienti, in linea con i principi delle 3R. L’integrazione della dieta con additivi funzionali può contribuire in modo significativo a ridurre l’insorgenza di patologie, migliorare la salute e la crescita degli animali, riducendo l’uso di antibiotici. L’uso di additivi funzionali si configura come un’interessante alternativa in linea con i principi di “One Health”.
