The ability of 25 compounds to antagonize the action of acetylcholine (ACh) at the muscarinic receptors mediating autoregulation of ACh release from cholinergic nerve terminals was comparatively studied using rat brain cortex synaptosomes in superfusion. The antagonistic activity at central cholinergic autoreceptors appears to depend strongly on the presence of a bulky basic moiety as well as on the way by which this moiety is connected to the aromatic portion of the molecule.
