In 21 anaesthetised and mechanically ventilated pigs we analysed the activity of 3-aminobenzamide (3-AB), a PARS (Poly ADP-ribose synthetase) inhibitor, in a model of septic shock induced by administration of E. Coli lipopolysaccharide (LPS). The evaluated parameters were: systemic (MAP) and pulmonaru (MPAP) arterial pressure, cardiac output (CO), systemic (SVR) and pulmonary (PVR) vascular resistances and compliance (Crs) and resistance (Rrs) of the respiratory system. Results show that 3-AB reduced the LPS-dependent bronchoconstriction, showing an involvement of PARS only at epithelial respiratory level, without involving vascular endothelium. Data related to exhaled NO evidenced that LPS did not enhance the NO release from lower airways.
