Spleen cell cultures from immunized and control mice have been followed in their morphological evolution, especially towards "blastoid" and binucleated cells. The immunological activity of cultures from immunized mice has been tested at different ages of culture by the Jerne antibody plaque essay. The ability of spleen cells to synthetize antibody was maintained in vitro for 15 days. DNA and RNA synthesis have been studied by radioautography using thymidine and uridine as precursors, respectively. No differences in the kinetic of synthesis appeared by relating the two types of cultures.
