Bovine neutrophils (PMNs) activated by incubation of blood samples in presence of lipopolysaccharides (LPS) showed changes in density levels respect to non activated samples, as previously observed under spontaneous inflammatory conditions. By the mean of discontinuous Percoll gradients it was possible to separate blood PMNs with very high yield and purity percentages at the highest Percoll concentration used. In activated samples the percentage of purity affects that of recover. The latter appears improved if the recover includes not only the cells that settle in correspondence of the highest Percoll concentration, but also those at the mean Percoll concentration, thus only slightly compromising purity levels. Flow cytometry demonstrated that activated PMNs are larger and show lower density and higher expression of integrins CD11b than non activated PMNs.
