A method has been developed for the determination of cholesterol in dairy products by high performance liquid chromatography. Saponification and extraction steps are completed rapidly in a single reaction tube, and a subsequent clean up is not required. Chromatographic separation requires 6 min. The within-batch precision of the HPLC method applied to butter presents a relative standard deviation of 1.6%. Extraction efficiency from the lipid matrix is approximately 100%. Reproducibility of the method, expressed as relative standard deviation, is less than 2%, and accuracy checked with a reference standard material (CBR164) is quite good.
