In the more popular cells employed for epithelial transport studies, all derived from a few early prototypes, the perfusing solution is circulated and aerated by means of air pumps in direct contact with the atmosphere. From the safety point of view, such a procedure cannot be recommended when radioactive tracers are employed. This also applies when the cells are operated in a filter hood as the filter pollution is increased, this being neither safe nor economical. An inherently safe, closed circulation, cell has been recently proposed, but no aeration was provided. Others have connected the air pump to a vacuum trap, but the perfusing fluid circulated only on one side of the membrane. In order to overcome these limitations, we have devised a new flow diagram.
