Micro RNAs are small non coding RNAs which play important regulatory roles at post trascritional level. They can bind by not complete base pairing to mRNA promoting degradation or blocking transcription. Mir-9 is a highly conserved miRNA expressed in developing nervous system of vertebrates and invertebrates. It is expressed at high level in some form of cancer, as in breast cancer, where it is supposed to promote metastasis by negative control of cadherins levels. We inhibited miR-9 function in Ciona intestinalis by microinjecting Peptide Nucleic Acid (PNA) antigomir in eggs. In PNA structure, the ribose phosphate backbone is replaced by N-(2-aminoethyl) glycine units, to form a pseudopeptide chain bearing the four nucleobases. PNAs show high selectivity and noticeable chemical and enzymatic stability. Larvae developed from microinjected eggs showed anomalies in the posterior region of neural tube and very few differentiated CENs, as showed by in situ hybridization with CNS and PNS markers. These results are similar to those obtained activating canonical Wnt signaling by LiCl treatments, suggesting an interaction between miR-9 activity and Wnt signaling in the differentiation of peripheral neurons. Instead, when the Notch pathway was blocked by incubation with DAPT, numerous scattered groups of cells were stained by both the markers. The immunofluorescence with anti-β-tubulin showed groups of 4-5 cells extending their cilia in the overlying tunic medial fin. These results suggest that multiple signals are involved, probably through different pathways, in controlling timing and balancement of CESNs differentiation.
