The aim of the following integrated project is to set up reporter gene assays based on primary neuronal cultures making use of identified neuronal specific response elements. This could represent a potential decision-making assay to be included into battery of tests for the characterization of novel antidepressants, as well as in the HTB screening battery. Towards this aim, we will take advantage of the recent knowledge of BDNF gene promoters and regulatory elements, as well as of the availability of a high-efficiency transfection system for neurons (Amaxa Nucleofection) which in-house led to approximately 40% transfection efficiency of primary hippocampal and cortical neurons.

Abstract
